I played Gladiator in OB, Templar in Live. Gladiators will do fine tanking if you can't find a Templar. Hell, I've seen Chanters tank instances. You won't be tanking Fortress Generals, but you can tank Training Grounds or Fire Temple just fine with the right support.
In regards to PvE tank, obviously Templar is superior, but honestly that's mostly only in terms of threat generation. The major difference defensively is Gladiators are limited to Shield Defense (Block+500, PhysDef+10%, StunRes+500) while Templars gain access to the upgraded Steel Wall Defense (Block+1000, PhysDef+15%, Stun/Knockback/Stumble/Spin/Aether's Hold Resist+1000, AtkSpeed-50%).
Passives: Templar: Boost HP V (15%) Boost Phys Atk III Boost Shield Def II Boost Element Def II Aggravation II (passive threat boost)
Gladiator: Boost HP III (9%) Boost Phys Atk V Boost Shield Def I Boost Knockdown Aggravation II (passive threat boost)
Gladiator's only real advantage in a tank role is their higher attack (knockdowns only happen with a 2H weapon), Templars have higher block rates (possibly*), bigger health pools, and some innate magic resistance. Glads also only have a single Provoke, they'll be holding threat through pure damage multiplied by their Aggravation bonus. Templars have Provoke as well as a Mocking Blow-like strike and a shield bash with a +threat modifier baseline, and they gain access to a Provoke, a Death Grip, and an AoE Provoke through Stigmas.
The reason I say possibly on block rates is that Aion handles avoidances in an interesting way. Evasion, Parry, and Block all gain 1% per 10 points of skill. It's good to note that as a Templar, my baseline block at Lv33 is around 1100. That translates into 110% blockrate. However. The game checks in the order of Evade > Parry > Block. Evade is capped at 30%, Parry is capped at 40%, and Block is capped at 50%. A Templar will likely be capped in block passively just by having a shield equipped. An Evade is 100% damage mitigated, Parry is a static 30% damage mitigated, Block varies from 25-60% mitigation based on your shield's quality and enchant level.
In terms of PvP, the addition to the above paragraph is that for each point of attacker's accuracy, all three (block, parry, and evade) of your avoidances are lowered by one point. It's not unusual to see rangers with over 1300 accuracy, effectively nullifying all of your avoidance.
As far as how the two classes play in PvP, they're worlds apart. Gladiators are purely offensive. They hit hard, knock enemies down a lot, have access to an Aether Hold, have a brutal AoE chain (that also procs knockdowns often), and have the plate armor to wade into a group of assassins and start wrecking faces. Their major weakness, however, is that they lack abilities to close in melee range. Any of the caster types will just kite them in circles.
Templars (they also play like this in PvE) rely heavily on reactive abilities. The few abilities that Templars have access to all the time don't hit very hard or are focused around defense. The most you'll be able to do is poke and harass healers, or use Inescapable Judgement (Death Grip + Chains of Ice + Stun on a 30 sec CD) to drag vulnerable enemies out into the open. Your goal as a Templar in PvP is to goad enemies into attacking you. As soon as an enemy Glad/Assassin/Chanter is dumb enough to try and melee you and you block an attack, all of your big-hitters become available as well as a couple of Stuns and Stumble-centric abilities. You -are- the front line in PvP scenarios. You cannot be killed if you have a healer watching you, unless you get too reckless or an extremely large number of ranged attackers all hit you at the same time (I'm talking 5-6 Sorcerers all using their big 2min CD nuke on you simultaneously). You're there to distract and dish out the pain to anyone stupid enough to attack you. Your weakness again is your vulnerability to kiting, but you have a larger selection of abilities that increase your resistances or remove debuffs from yourself, as well as Inescapable Judgement.
